GHRP-6 was developed by Bowers from met-enkephalin analogues. Its activity at an unidentified receptor prompted the expression-cloning work that identified GHS-R1a, and later the search for the endogenous ligand that produced ghrelin. It remains the reference agonist in ghrelin-receptor pharmacology.
