Long R3 IGF-1 carries a glutamate-to-arginine substitution at position 3 and a 13-residue N-terminal extension. Both modifications sharply lower affinity for the IGF-binding proteins, which is why the analogue is the standard IGF-1 receptor agonist in serum-free cell culture and bioprocess media literature.
